1.PBI buffer provides the optimal salt concentration and pH for adsorption of high molecular weight DNA to the membrane.DNA adsorption to the silica-gel surface also requires a high
concentration of a "chaotropic agent" such as guanidine hydrochloride.
2.Buffer PE efficiently washes away contaminants.During the DNA adsorption step,unwanted contaminants such as primers,salts,enzymes,and unincorporated nucleotides do not bind to the silica membrane,but flow through the column.
3.Buffer QG has 2 functions,1).solubilize the agarose gel slice; 2).provide the appropriate conditions for binding of DNA to the silica membrane,pH
